X-Cell detection systems are designed for reliable, cost-effective, two-step detection, providing a high-level of sensitivity. The vast majority of primary antibodies can be diluted two-fold compared to other commercially available avidin-biotin detection systems. MenaPath prediluted antibodies are all standardised to work with the X-Cell detection system.

X-Cell detection system can be used with mouse and rabbit primary antibodies. After labelling the antigen with primary antibody, a species-specific or universal, affinity-purified, biotinylated secondary antibody is added to bind. Horseradish peroxidase (HRP) or alkaline phosphatase (AP) labelled-streptavidin is then added. A chromogen/substrate is applied that reacts with a HRP or AP to produce an intense colour signal.

MenaPath Avidin-Biotin Blocking Kits are optimised for blocking endogenous biotin in tissue sections. Endogenous biotin is abundant in cells such as hepatocytes, kidney tubules and many tumours, particularly carcinomas. MenaPath biotin-blocking kits can eliminate nonspecific false-positive staining when using avidin-biotin detection systems. The blocking step is particularly critical when using heat-induced epitope retrieval (HIER), which unmasks endogeneous biotin in tissues.

Streptavidin is a protein that has similar binding properties to egg white avidin. It is isolated from streptomyces avidinii. Streptavidin has a molecular weight of 60 kDa and has 4 subunits. Each subunit can bind one molecule of biotin. Biotin is a water- soluble vitamin. Streptavidin has an extremely high binding affinity (Kd=10-15) for biotin. Streptavidin conjugated to alkaline phosphatase has proven useful in the detection of antigens coupled with biotinylated secondary antibodies. There are several advantages when using a streptavidin-conjugate versus an ABC complex.In contrast to avidin, streptavidin is not glycosylated and is therefore uncharged at neutral pH (6.5 versus 10). This lowers nonspecific background staining. Streptavidin also lacks carbohydrate side chains that may be another cause of non-specific background. Another key advantage of streptavidin is the significant increase in sensitivity (probably due to less steric hindrance), thus facilitating an increase in overall binding capacity. Streptavidin-AP is much more stable than the ABC complex. The ABC complex must be freshly made 30 minutes prior to use, and is stable only for a few days. In contrast, the Streptavidin-AP reagent is very stable, colour-coded, and can be stored for up to 1-2 years. This Streptavidin-AP comes in a ready-to-use format, thus saving time and potential mistakes.

Mouse monoclonal antibodies can be used for immunohistochemical detection on mouse tissues; however, the biotinylated secondary antibody used for detection of the primary antibody will also bind to the endogenous mouse IgG in the tissue, resulting in background staining from bound-mouse IgG. To circumvent the problem,  please use the MM Biotinylation Kit. The kit provides all the reagents necessary to biotinylate a primary antibody.

The basic concept of the kit is very simple. The antibody is mixed with a biotinylation reagent for 30 minutes prior to the application of primary antibody. As little as 5 to 10 µL of antibody can be used. Once the complex is formed, a mopping reagent is added and then the biotinylated antibody complex is applied to the tissue specimen. The tissue is then incubated with the strepavidin-peroxidase reagent, and the colour reaction is developed by a DAB chromogen substrate.

The system has been specially formulated to eliminate endogenous mouse IgG background staining. Even the most difficult tissues such as lymph, spleen, lung and kidney are virtually background free.
